CDK4/6 inhibitors stall the cell cycle. By blocking cyclin-dependent kinases 4 and 6, they stop breast cancer cells from moving out of the growth phase and dividing. They are given as a daily tablet alongside hormone therapy in hormone-receptor-positive, HER2-negative advanced breast cancer, where adding them roughly doubles the time before the disease progresses.
