BTK inhibitors block Bruton's tyrosine kinase, an enzyme that B-cells depend on to survive and multiply. Shutting it down starves B-cell cancers such as chronic lymphocytic leukaemia and mantle cell lymphoma. Ibrutinib was first; the newer, more selective agents were developed to hit BTK harder while touching fewer off-target kinases, which changes their side-effect profile.
